People rarely have HSV resistant to acyclovir – the rate is 0.3% of strains are resistant. And if one did have a resistant strain, it would still be antibody positive.
I have not seen herpes proctitis but it has been documented, yes, and it would subside in days or weeks after the outbreak.
If you met with me, I would evaluate pictures. The western blot is about $450 total.
I think you are totally barking up the wrong tree on this diagnosis
